U-PLEX Custom Multiplex Assay Configuration
Traditional custom multiplex assay development can take months and cost tens of thousands of dollars — antibody pairs must be screened for cross-reactivity, spot conditions optimized, and the full panel validated. U-PLEX eliminates this timeline through a modular, linker-based architecture. Each target antibody is pre-coupled to a unique DNA-based linker that directs it to a specific spot on the MSD plate. Since the antibodies are individually qualified and the linker-spot assignment is deterministic, assembling a custom panel is a configuration exercise — not a development project.
When U-PLEX Is the Right Choice
- Your target list doesn't match any pre-configured V-PLEX panel: You need 7 specific analytes spread across three different V-PLEX panels — U-PLEX consolidates them into one assay.
- You're adding exploratory targets to a validated core: Run a V-PLEX panel for your primary endpoints and a U-PLEX panel for secondary/exploratory targets — all from the same sample.
- Budget or sample volume constraints favor a focused panel: You only need 5 specific cytokines — a 54-plex is overkill. U-PLEX lets you measure exactly what you need, nothing more.
- You need species-specific targets not available in pre-configured panels: U-PLEX can draw from a broader catalog including targets validated for mouse, rat, and NHP that may not be available in fixed V-PLEX configurations.
Panel Design Process
- Submit your target list — send us the analytes, species, and matrix you plan to use. We verify target availability in the U-PLEX catalog and flag any targets that require alternative sourcing.
- Compatibility review — our scientific team evaluates your combination for known interactions, concentration range compatibility, and matrix performance based on existing U-PLEX data for each individual target.
- Feasibility confirmation — we confirm the panel can be assembled and provide a project timeline. Most U-PLEX panels ship within 2–3 weeks of order confirmation.
- Assay execution — plates are prepared with your custom linker-coupled antibody set. Standard curves, QC samples, and acceptance criteria follow the same framework as V-PLEX assays.
U-PLEX Catalog Scope
The U-PLEX Human Biomarker Group 1 catalog covers 50+ individually qualified targets spanning cytokines (IL-1α through IL-33, 20+ interleukins), chemokines (Eotaxin family, IP-10, MCP-1/2/3/4, MIP-1α/1β/3α/3β/5, MIG, I-TAC, SDF-1α, CTACK, Fractalkine), growth factors (VEGF-A, GM-CSF, G-CSF, M-CSF), soluble receptors, and acute-phase proteins. Mouse, rat, and NHP catalogs provide species-matched coverage for translational studies. Targets not in the current catalog can be evaluated for R-PLEX custom development. Contact us with your analyte list — we'll confirm catalog availability within 1–2 business days.
Custom Panel Configuration — Autoimmune Research Application
A researcher studying psoriatic arthritis needed to measure five analytes not covered by any single V-PLEX panel: IL-17A (Th17 effector), IL-23 (Th17 polarizing), TNF-α (inflammatory), IP-10 (Th1 chemokine), and MIP-3α (CCR6 ligand — Th17 recruitment). These targets were spread across three different V-PLEX panels. Via U-PLEX, all five were combined into a single well, consuming 25 µL per sample instead of the 75 µL that three separate panels would require. The custom panel was assembled and verified within two weeks — no custom development timeline, no minimum order beyond the five-analyte configuration.
