Applications Requiring Ultrasensitive Cytokine Detection
Immunoassay sensitivity is conventionally reported as the lower limit of detection (LOD) — the lowest concentration statistically distinguishable from zero. But the practical metric that matters for research is the lower limit of quantification (LLOQ) — the lowest concentration that can be measured with acceptable precision (CV <20%). The gap between LOD and LLOQ is where biologically meaningful cytokine signals reside, and where standard assays fall short.
Scenarios Where S-PLEX Matters
- Baseline cytokine levels in healthy populations: IL-4, IL-5, IL-13, and other Th2 cytokines typically circulate at <1 pg/mL in plasma — below the LLOQ of standard multiplex assays. S-PLEX brings these into the quantifiable range, enabling group comparisons that standard assays cannot support.
- Early treatment effects: When an investigational therapy suppresses an already low-abundance cytokine, standard assays may report "below LLOQ" for both pre- and post-treatment samples — masking a real biological effect. S-PLEX provides the resolution to detect these changes.
- Precious sample volumes: Pediatric samples, CSF, longitudinal aliquots nearing depletion — when sample volume is limiting, the assay must extract maximum information from every microliter. S-PLEX requires only 25 µL per well while delivering the highest sensitivity available on the MSD platform.
- Biomarker discovery in early disease: The earliest cytokine perturbations in disease may occur at concentrations far below those seen in established pathology. S-PLEX extends the measurable range downward, potentially revealing signals missed by standard-sensitivity approaches.
